  • 제목: Gene Expression Programs Underlying Synchronous Network Activity

  • Abstract:

    Neurons commonly fire in synchrony, a phenomenon critical for cognitive function. This coordinated firing requires activity-dependent fine-tuning of neuronal connectivity and excitability, processes in which gene expression plays a central role. However, the gene expression programs that support coordinated firing remain largely unknown. To address this, we developed CalTRAP-seq, a method for profiling gene expression in active neurons via calcium-dependent ribosome tagging. Applying CalTRAP-seq to active neuronal networks revealed distinct gene expression patterns associated with spontaneous and synchronous network activity, characterized by extensive regulation of neuronal excitability. Furthermore, we found that neuronal activity promotes nuclear speckle formation and changes alternative splicing of numerous synaptic genes. Disrupting nuclear speckles impaired the dynamics of network activity. Together, these findings identify a novel gene expression program linked to spontaneous network activity, providing new insight into how neurons regulate gene expression to support coherent network function.
